Joe Worsley tore medial ligaments in his right knee in the opening minute of England's 19-6 defeat to New Zealand on Saturday.
The Wasps flanker went down awkwardly in a tackle and had to be replaced by Leicester's Tom Croft.
Worsley, who had only just returned to the England team following a neck injury, is to undergo a scan over the next 24 hours which will determine how long he will be sidelined for.
The Rugby Football Union announced in a statement: "Further to an assessment and review by the England medical team and consultation with the London Wasps medics this morning it can be confirmed that Joe Worsley tore medial ligaments in his right knee during the Investec Challenge England v New Zealand Test match.
"Joe will have a scan over the next 24 hours. Once this has been completed a further announcement will be made regarding the length of time that Joe will be unavailable."
